    The effect of BMP9 on inflammation in the early stage of pulpitis

    Bone morphogenetic protein 9 (BMP9) tends to be associated with various inflammatory responses of diseases, but its relationship with pulpitis remains unknown. Objective: This study aimed to evaluate the effects and mechanisms of BMP9 in pulpitis. Methodology: A rat model of pulpitis was used to evaluate the expression of BMP9, which was also analysed in Porphyromonas gingivalis lipopolysaccharide (Pg-LPS)-stimulated human dental pulp cells (hDPCs). The effects and mechanism of BMP9 on the regulation of inflammatory factors and matrix metalloproteinase-2 (MMP2) were evaluated using real-time quantitative PCR, western blotting, and immunocytofluorescence. Moreover, the migration ability of THP-1 monocyte-macrophages, treated with inflammatory supernate inhibited by BMP9, was previously tested by a transwell migration assay. Finally, a direct rat pulp capping model was used to evaluate in vivo the influence of the overexpression of BMP9 in pulpitis. Results: The expression of BMP9 decreased after 24 h and increased after 3 and 7 d in rat pulpitis and inflammatory hDPCs. The overexpression of BMP9 inhibited the gene expression of inflammatory factors (IL-6, IL-8, and CCL2) and the secretion of IL-6 and MMP2 in Pg-LPS-stimulated hDPCs. The level of phosphorylated Smad1/5 was upregulated and the levels of phosphorylated ERK and JNK were downregulated. The inflammatory supernate of hDPCs inhibited by BMP9 reduced the migration of THP-1 cells. In rat pulp capping models, overexpressed BMP9 could partially restrain the development of dental pulp inflammation. Conclusion: This is the first study to confirm that BMP9 is involved in the occurrence and development of pulpitis and can partially inhibit its severity in the early stage. These findings provided a theoretical reference for future studies on the mechanism of pulpitis and application of bioactive molecules in vital pulp therapy

    NMR determination of Van Hove singularity and Lifshitz transitions in nodal-line semimetal ZrSiTe

    We have applied nuclear magnetic resonance spectroscopy to study the distinctive network of nodal lines in the Dirac semimetal ZrSiTe. The low-TT behavior is dominated by a symmetry-protected nodal line, with NMR providing a sensitive probe of the diamagnetic response of the associated carriers. A sharp low-TT minimum in NMR shift and (T1T)āˆ’1(T_1T)^{-1} provides a quantitative measure of the dispersionless, quasi-2D behavior of this nodal line. We also identify a van Hove singularity closely connected to this nodal line, and an associated TT-induced Lifshitz transition. A disconnect in the NMR shift and line width at this temperature indicates the change in electronic behavior associated with this topological change. These features have an orientation-dependent behavior indicating a field-dependent scaling of the associated band energies.Comment: 7 pages, 4 figure

    Giant room temperature anomalous Hall effect and magnetically tuned topology in the ferromagnetic Weyl semimetal Co2MnAl

    Weyl semimetals (WSM) have been extensively studied due to their exotic properties such as topological surface states and anomalous transport phenomena. Their band structure topology is usually predetermined by material parameters and can hardly be manipulated once the material is formed. Their unique transport properties appear usually at very low temperature, which sets challenges for practical device applications. In this work, we demonstrate a way to modify the band topology via a weak magnetic field in a ferromagnetic topological semimetal, Co2MnAl, at room temperature. We observe a tunable, giant anomalous Hall effect, which is induced by the transition between Weyl points and nodal rings as rotating the magnetization axis. The anomalous Hall conductivity is as large as that of a 3D quantum anomalous Hall effect (QAHE), with the Hall angle reaching a record value (21%) at the room temperature among magnetic conductors. Furthermore, we propose a material recipe to generate the giant anomalous Hall effect by gaping nodal rings without requiring the existence of Weyl points. Our work reveals an ideal intrinsically magnetic platform to explore the interplay between magnetic dynamics and topological physics for the development of a new generation of spintronic devices.Comment: 4 figures, 8 pages for the main text.     Derivation of hypermethylated pluripotent embryonic stem cells with high potency.

    Naive hypomethylated embryonic pluripotent stem cells (ESCs) are developmentally closest to the preimplantation epiblast of blastocysts, with the potential to contribute to all embryonic tissues and the germline, excepting the extra-embryonic tissues in chimeric embryos. By contrast, epiblast stem cells (EpiSCs) resembling postimplantation epiblast are relatively more methylated and show a limited potential for chimerism. Here, for the first time, we reveal advanced pluripotent stem cells (ASCs), which are developmentally beyond the pluripotent cells in the inner cell mass but with higher potency than EpiSCs. Accordingly, a single ASC contributes very efficiently to the fetus, germline, yolk sac and the placental labyrinth in chimeras. Since they are developmentally more advanced, ASCs do not contribute to the trophoblast. ASCs were derived from blastocysts in two steps in a chemically defined medium supplemented with Activin A and basic fibroblast growth factor, followed by culturing in ABCL medium containing ActA, BMP4, CHIR99021 and leukemia inhibitory factor. Notably, ASCs exhibit a distinct transcriptome with the expression of both naive pluripotency genes, as well as mesodermal somatic genes; Eomes, Eras, Tdgf1, Evx1, hand1, Wnt5a and distinct repetitive elements. Conversion of established ESCs to ASCs is also achievable. Importantly, ASCs exhibit a stable hypermethylated epigenome and mostly intact imprints as compared to the hypomethylated inner cell mass of blastocysts and naive ESCs.     Adenovirus-delivered CIAPIN1 small interfering RNA inhibits HCC growth in vitro and in vivo

    Hepatocellular carcinoma (HCC) is an aggressive cancer with a poor prognosis. The specific cellular gene alterations responsible for hepatocarcinogenesis are not well known. Cytokine-induced antiapoptotic molecule (CIAPIN1), a recently reported antiapoptotic molecule which plays an essential role in mouse definitive hematopoiesis, is considered a downstream effecter of the receptor tyrosine kinaseā€“Ras signaling pathway. However, the exact function of this gene in tumors is not clear. In this study, we reported that CIAPIN1 is highly expressed in HCC as compared with non-tumor hepatic tissue (P < 0.05). We employed adenovirus-mediated RNA interference technique to knock down CIAPIN1 expression in HCC cells and observed its effects on HCC cell growth in vitro and in vivo. Among the four HCC and one normal human liver cell lines we analyzed, CIAPIN1 was highly expressed in HCC cells. Knock down of CIAPIN1 could inhibit HCC cell proliferation by inhibiting the cell cycle S-phase entry. Soft agar colony formation assay indicated that the colony-forming ability of SMMC-7721 cells decreased by āˆ¼70% after adenovirus AdH1-small interfering RNA (siRNA)/CIAPIN1 infection. In vivo experiments showed that adenovirus AdH1-siRNA/CIAPIN1 inhibited the tumorigenicity of SMMC-7721 cells and significantly suppressed tumor growth when injected directly into tumors. These results suggest that knock down of CIAPIN1 by adenovirus-delivered siRNA may be a potential therapeutic strategy for treatment of HCC in which CIAPIN1 is overexpressed

    MiR-218 Inhibits Invasion and Metastasis of Gastric Cancer by Targeting the Robo1 Receptor

    MicroRNAs play key roles in tumor metastasis. Here, we describe the regulation and function of miR-218 in gastric cancer (GC) metastasis. miR-218 expression is decreased along with the expression of one of its host genes, Slit3 in metastatic GC. However, Robo1, one of several Slit receptors, is negatively regulated by miR-218, thus establishing a negative feedback loop. Decreased miR-218 levels eliminate Robo1 repression, which activates the Slit-Robo1 pathway through the interaction between Robo1 and Slit2, thus triggering tumor metastasis. The restoration of miR-218 suppresses Robo1 expression and inhibits tumor cell invasion and metastasis in vitro and in vivo. Taken together, our results describe a Slit-miR-218-Robo1 regulatory circuit whose disruption may contribute to GC metastasis. Targeting miR-218 may provide a strategy for blocking tumor metastasis

    Aiming at the problem that the traditional MART algorithm has low iteration accuracy,a new tomographic inversion algorithm is proposed. On the one hand,The new algorithm takes into account the integration of the intercept of ray traversing pixels and electron density and distributes the iterative difference in a more reasonably. On the other hand,an adaptive relaxation factor related to the electron density is proposed to overcome the influence of propagation noise on electron density inversion. The GIM data and GPS dual frequency observation data are used to verify the feasibility and superiority of the new method iteration accuracy from two aspects of single ray iteration and multi ray iteration. The experimental results show that compared with the traditional MART algorithm,the electron density profile obtained by the new method is closer with those from ionosonde measurements

    Bird activities including the defecation, nesting, and flight are easy to cause electrical faults of overhead transmission lines, study on the prevention measures of bird damage to transmission lines is of great significance to ensure the stable and reliable operation of the power system. With the establishment of finite element simulation model, the effects of box of bird guard with different material on the electric field and potential distribution on transmission line insulators are studied. Besides, combined with the theory of air discharge and the results of simulation, the influence of rainwater on the air gap around the insulator breakdown characteristics is studied when the water is drained from a scupper at the bottom of the box of bird guard, and a reasonable design scheme for the box of bird guard scupper is proposed. The research results here can provide a guidance for the selection and key parametersā€™ design of the box of bird guard
